Nearly 200 golfers are expected to attend and will be split into 36 groups of five, which will each be paired with a celebrity.
Shaquille O'Neal, Bobby Knight, Mike Piazza and Bo Jackson have attended the event as celebrity guests in the past.
Proceeds from the event will benefit the Pujols Family Foundation to assist orphans in the Dominican Republic, in addition to those with Down syndrome.
